赤水金钗石斛"鱼肚兰"的基因组特征

[目的]探明赤水金钗石斛"鱼肚兰"野生种与栽培种的基因组特征,为赤水金钗石斛的资源保护及品种选育提供遗传信息支撑.[方法]采用高通量测序技术、显微观察等方法研究赤水金钗石斛"鱼肚兰"的野生种与栽培种,并分析其基因组大小、数量、倍性等特征.[结果]高通量测序共获得赤水金钗石斛"鱼肚兰"野生种和栽培种有效序列长度分别 53.30 Gb和 60.62 Gb,k-mer分析基因组估测的基因组大小分别为 1.18 Gb和 1.13 Gb,重复序列比例分别为 50.56%和 57.59%,基因组杂合度分别为 2.19%和 2.76%;根尖染色体数目的显微计数显示,野生种与栽培种赤水金钗石斛均为二倍体(2n=2x=38),与基因组调研结果一致.[结论]赤水金钗石斛"鱼肚兰"的野生种与栽培种均为二倍体,从基因组特征上看,二者均属于高杂合的复杂基因组.
Genomic Characteristics of Chishui Dendrobium nobile'Yudulan'
[Objective]The genomic characteristics of wild and cultivated Chishui Dendrobium nobile'Yudulan'were explored,which provided genetic information support for resource conservation and breed selection of Chishui D.nobile.[Method]High-throughput sequencing technology and microscopic observation were used to study the genome size,quantity and ploidy of wild and cultivated species of Chishui D.nobile'Yudulan'.[Result]The effective sequence length of wild and cultivated Chishui D.nobile were respectively 53.30 Gb and 60.62 Gb by high-throughput sequencing technology.The genome size estimated by k-mer analysis was 1.18 Gb and 1.13 Gb,and the proportion of repeat sequences was 50.56%and 57.59%.The genome heterozygosity was 2.19%and 2.76%,respectively.The microscopic counting results of the number of chromosomes at the root tip showed that both wild and cultivated Chishui D.nobile species were diploid(2n=2x=38),which were consistent with the results of genomic investigation.[Conclusion]Both wild species and cultivars of Chishui D.nobile are diploid,and both belong to complex genomes with high heterozygosity from the perspective of genomic characteristics.
