Characterization Effects of"Drumstick-forming"on Amino Acid and Dencichine Contents in Medicinal Parts of Panax notoginseng
[Objective]The effects of the top stem segment thickening like chicken leg in the above-ground part of Panax notoginseng(commonly known as"drumstick-forming")on the amino acid and dencichine contents in the traditional medicinal parts of P.notoginseng were explored to provide the theoretical basis for the good-quality and high-yield of P.notoginseng.[Method]The total free amino acid contents(TFAC)and dencichine content(DC)in the taproots, lateral roots, rhizomes and underground organs of P.notoginseng plants with or without"drumstick-forming"were determined by using ninhydrin method and high performance liquid chromatography, respectively. On this basis, the variance analysis was used to study the characterization effects of"drumstick-forming"on the amino acid and dencichine contents in the traditional medicinal parts of P.notoginseng.[Result]Compared with"non-drumstick-forming"plants,for the 2-year-old"drumstick-forming"ones,TFAC in the taproot lowered with 18.92%at a non-significant level,TFAC in the rhizome and underground organ lowered with 31.06%and 16.26%respectively at significant levels, whereas TFAC in the lateral root increased by 14.26%at a significant level.DC in the taproot, lateral root and underground organ lowered with 33.87%,26.55%and 17.97%respectively at extremely significant levels, whereas DC in the rhizome increased by 9.37%at an extremely significant level. For the 3-year-old"drumstick-forming"ones,TFAC in the taproot increased by 13.90%at a significant level, whereas TFAC in the lateral root, rhizome and underground organ lowered with 1.13%,14.32%and 1.45%respectively at non-significant levels.DC in the taproot and rhizome lowered with 9.96%and 8.61%respectively at significant levels, whereas DC in the lateral root increased by 18.14%at a significant level and DC in the underground organ increased by 0.37%at a non-significant level.Meanwhile, from 2-to 3-year old,TFAC in the lateral root, rhizome and underground organ of the"non-drumstick-forming"and"drumstick-forming"plants all decreased, and the latter decreased by 673.98%,-31.21%and-44.82%compared to the former, respectively, whereas DC in the taproot both increased, and the latter increased by 5.74 times compared to the former.[Conclusion]The"drumstick-forming"characterizes the higher TFAC of the 2-year-old lateral roots and 3-year-old taproots, the higher DC of the 2-year-old rhizomes,3-year-old lateral roots and underground organs, and the increases of TFAC in the taproot and DC in the taproot, lateral root and underground organ from 2-to 3-year old. However, the TFAC and DC of other medicinal parts with different growth years are lower, and from 2-to 3-year old,the TFAC and DC in different parts all decrease.
