Optimization of Ultrasonic-assisted Extraction of Crude Polysaccharides from Epimedium brevicornu by Box-Behnken Response Surface Method
[Objective]The optimal technique for extracting crude polysaccharide of Epimedium brevicornu by ultrasonic-assisted extraction method was studied,which provided basis for extracting and utilizing polysaccharide in the medicinal plant E.brevicornu.[Method]Taking E.brevicornu as the study object,the ultrasonic-assisted extraction method combined with the single factor test to screen the optimal extraction temperature,material to liquid ratio and extraction time of polysaccharide in E.brevicornu.On the basis,the optimized process parameters were determined as the extraction of crude polysaccharide in E.brevicornu by the response surface method.[Result]The optimal extraction parameters of crude polysaccharide in E.brevicornu were extraction temperature of 60℃,material to liquid ratio of 1∶20(g∶mL)and extraction time of 40 min.Under the conditions the extraction rate was 7.47%,which was basically consistent with the theoretical value(7.57%).The relative standard deviation(RSD)of repeated tests was 0.65%,which met the test requirements.[Conclusion]The extraction process of crude polysaccharide from E.brevicornu is optimized by the response surface method,which can extract crude polysaccharide from E.brevicornu rapidly and efficiently,and the process has the stable extraction rate.It lay a foundation for better exploitation of crude polysaccharide resources in E.brevicornu in the future.
