Genetic Diversity and DNA Fingerprint Construction for 50 Dendrobium Species
[Objective]The genetic diversity of Dendrobium germplasm resources introduced domestically and internationally was analyzed to uncover the genetic relationship among different germplasms,and to provide a reference for promoting effective utilization and preservation of Dendrobium germplasm.[Method]The genetic diversity and genetic relationship among 50 Dendrobium species,including Dendrobium gratiosissimum,D.nobile,D.anosmum,etc.,were analyzed by ISSR molecular marker technology,as well as a DNA fingerprint was constructed.[Result]9 primers with high polymorphism and good repeatability amplified a total of 112 alleles,including 102 polymorphism sites and a polymorphism ratio of 93.09%.The number of detected alleles ranged from 9 to 14,with an average of 12.44.The genetic similarity coefficients among the 50 Dendrobium species ranged from 0.15 to 0.95.The cluster analysis revealed that the 50 Dendrobium species could be divided into 12 groups at a genetic similarity coefficient of 0.70.PCR amplification showed that primer UBC808 successfully distinguished all 50 Dendrobium species.Additionally,DNA fingerprint of the 50 Dendrobium species was constructed based on 14 clear polymorphic bands amplified with this primer.[Conclusion]The 50 Dendrobium species have abundant genetic diversity.
