Study on the Identification and Biological Characteristics of a Wild Oudemansiella mucida
[Objective]A wild fungal strain was collected in Xizang area during 2021-2022 and numbered X21198,which laid the foundation for the subsequent development and utilization of the strain.[Method]The species characteristics of wild strain X21198 were determined by using morphological identification and ITS identification analysis,and the biological characteristics of mycelia were studied under the conditions of different carbon source(glucose,sucrose,fructose,maltose,mannose and starch),nitrogen source(urea,yeast powder,ammonium sulfate,beef meal,peptone and ammonium nitrate),pH(5.0,6.0,7.0,8.0,9.0 and 10.0)and temperature(15℃,20℃,25℃,30℃,35℃and 40℃).[Result]The cap of wild strain X21198 was hemispherical to flat,slimy or sticky,white and edge with sparse but not distinct stripes.The gill was straight to curved,sparse and unequal long.The stipe was cylindrical,solid and fibrous.The context was thin and white.The similarity of ITS sequence between X21198 and LC149596.1 Mucidula and OP328319.1 was 84%,belonging to the same group,and then the strain is identified as Oudemansiella mucida.The optimal carbon source,nitrogen source,pH and temperature was respectively fructose,yeast powder,8 and 25℃.[Conclusion]The wild strain X21198 in Xizang area was identified as O.mucida.The optimal carbon source,nitrogen source,pH and temperature for the mycelia culture is respectively fructose,yeast powder,8 and 25℃.
