Objective:To explore the effect of antigen retrieval time on the immunohistochemical staining results of P40 with different Clones.Methods:The pleural effusion cell sedimen with abnormal immunohistochemical staining of P40 was collected.Four sections of cell sediment with a thickness of 3 microns were divided into two groups according to antigen retrieval time.group A had a duration of 40 minutes and group B had a duration of 80 minutes.Performed immunohistochemical staining and added P40 antibodies of Clone C3B4 and MXR010 to two slices separately in each group.Results:In group A,C3B4 showed moderatenonspecific staining of cytoplasm and membrane,while MXR010 did not show nonspecific staining.In group B,C3B4 showed severe nonspecific staining of cytoplasm and membrane,while MXR010 showed mildnonspecific staining of cytoplasm and membrane.Compared the same clone between group A and group B,with the prolongation of antigen retrieval time,MXR010 changed from no nonspecific staining to mild nonspecific staining,while C3B4 changed from moderate nonspecific staining to severe nonspecific staining.Conclusion:The antigen retrieval time will have an effect on the immunohistochemical staining results of P40 with different clones.As the antigen retrieval time prolongs,nonspecific staining becomes more likely to occur.Choosing appropriate antigen retrieval time and antibodies with good specificity can reduce nonspecific staining.
