Study on Optimization of Ultrasonic Extraction Process and Antioxidant Activity of Soybean Saponins
In this study,the commercial domestic soybeans were used as raw materials to study the optimal ultrasonic ex-traction conditions of soybean saponin and its antioxidant activity.Based on the analysis of single factor effect test,the ex-traction rate of soybean saponin was used as the index to optimize the response surface analysis of the factors affecting the liquid-solid ratio,ultrasonic extraction time and ethanol volume percentage of soybean saponin extraction.The optimal pro-cess conditions for ultrasonic extraction of soybean saponin were obtained,and the ability of soybean saponin to remove DPPH free radicals and ABTS free radicals was discussed.The results showed that the optimum ultrasonic extraction condi-tions of soybean saponins were as follows:liquid-solid ratio was 45∶1(mL∶g),ultrasonic extraction time was 40 min,and ethanol volume percentage was 66%.Under the optimal ultrasonic extraction conditions,the average yield of soybean sapo-nins obtained by three parallel experiments was(4.21±0.13)mg/g,which was basically consistent with the predicted value of 4.22 mg/g by response surface optimization analysis.The IC50 values of soybean saponins against DPPH and ABTS free radicals were(0.44±0.06)mg/mL and(0.39±0.07)mg/mL,respectively.The results showed that the optimized ultrasonic extraction process parameters of soybean saponins were reliable.The extracted soybean saponins had ideal antioxidant activity,which could be used as a natural antioxidant in food production and feed processing.The research results pro-vide a theoretical basis for the industrial extraction of soybean saponins and the deep development and utilization of soy-bean resources.
