Study on the Mechanism of Improving Oxidative Stress and Insulin Resistance of HepG2 Cells by Using Nrf2/ARE Signaling Pathway
The main symptoms of type 2 diabetes include insulin resistance,high blood sugar and insulin deficiency.Al-though some oral hypoglycemic drugs can be effective in improving insulin resistance,most of them,including biguanides,thiazolidinediones,and sulfonylureas,cause serious adverse reactions,which has led to an interest in naturally sourced drugs to treat type 2 diabetes.In this study,the mechanism of the improvement of oxidative stress and insulin resistance of HepG2 cells by water extract of Yangxiensis based on Nrf2/ARE signaling pathway was investigated.After treatment with different concentrations of water extract of Yangxiensis,superoxide dismutase and glutathione peroxidase significantly in-creased,and the levels of reactive oxygen species and lipid malondialdehyde oxide significantly reversed.Meanwhile,the levels of nuclear factor erythrocyte-related factor 2,Kelch-like epichlorohydrin-related protein and heme oxygenase-1 protein significantly increased in insulin-resistant HepG2 cells.And the gene expressions of nuclear factor E2 related fac-tor 2,heme oxygenase 1 and insulin receptor substrate 1 significantly increased,which further confirmed that the water ex-tract of Yangzi can improve glucose metabolism,insulin resistance and oxidative stress of HepG2 cells induced by high glu-cose resistance.This study identified the potential of the water extract of Herba japonica to improve oxidative stress and in-sulin resistance and treat type 2 diabetes by activating Nrf2/ARE signaling pathway.
