不同树脂对胸腺素α1合成工艺的影响

摘要
目的:采用固相法合成胸腺素α1及其片段,比较二氯三苯甲基树脂、Wang树脂和Rink AmideMBHA树脂作为载体时对不同长度片段及目标肽的产率和纯度的影响.方法:FMOC策略合成胸腺素α1及其C端7肽、14肽、21肽片段,以反相高效液相色谱进行粗肽的分析和纯化.结果:氨基酸数小于10时,3种树脂合成的粗肽纯度均>95%,随着氨基酸数的增加,3种树脂合成的粗肽纯度均下降,其中Wang合成的粗肽纯度变化最明显,28肽纯度为21.24%.3种树脂合成粗肽1次纯化后,得到Tal纯度均大于95%;Wang树脂合成产率最高,CTC树脂产率最高.结论:以二氯三苯甲基树脂作载体,合成胸腺素α1其产率和纯度优于Wang树脂和Rink树脂,因此,二氯三苯甲基树脂作载体合成胸腺素α1在大规模工业化生产中具有更大的潜力.
Abstract
Objective: To explore the effect of 2-chlorotrity chloride resin, Wang resin and Rink Amide MBHA resin on the yield and purity of polypeptides by FMOC solid-phase method. Methods: Thymosin al and its various peptide fragments were synthesized by FMOC-based solid phased synthesis. The field and purity were analyzed by RP-HPLC. Results: The crude peptide purities of three resins were all lower than 95% when the number of amino acid was less than 10. The purity decreased as the amino acid increasing. And peptide of Wang showed the most significant decrease. The purity of peptide 28 was 21.24%. Tal from three resins all had purity higher than 21.24%. Wang resin produced the highest field,and CTC got the lowest. Oonclusion: The results demonstrate that the purity and yield of the peptide by using 2-chlorotrity choloride resin is higher than using Wang resin and Rink Amide MBHA resin. Long chain peptide can be synthesized with 2-chlorotrity choloride resin in large-scale.
