A study on the mechanism of action of Simiaoyong'an Decoction on adipocyte inflammation based on PPARγ/NF-κB signaling pathway
Objective:To investigate the effect of Simiaoyong'an Decoction on lipopolysaccharide(LPS)-induced inflammation in adipocytes and its underlying mechanism through the PPARγ/NF-κB signaling pathway.Methods:An adipocyte inflammation model was established by inducing mature adipocytes with LPS.The expression of proteins involved in the adipocyte inflammatory signaling pathway was evaluated after 24 h of intervention by administration of freeze-dried powder of Simiaoyong'an Decoction.The effect of Simiaoyong'an Decoction on adipocytes activity was detected by CCK-8 assay.The expression of IL-6,NLRP3,PPARγ,NF-κB p65,LXRα and GLUT4 by Western blot(WB).Results:CCK-8 results showed that freeze-dried powder of Simiaoyong'an Decoction had no significant effect on the viability of adipocytes in the range of 0~500 μg/mL.The WB results re-vealed that following LPS intervention,the model group exhibited increased intracellular inflammatory factors IL-6 and NLRP3 expression,decreased PPARγ expression,increased NF-κB p65 expression,and decreased LXRα and GLUT4 expression com-pared to the control group.Compared with the model group,Simiaoyong'an Decoction dose-dependently reduced IL-6 and NL-RP3 expression.Additionally,the expression of PPARγ,LXRα and GLUT4 increased,while the expression of NF-κB de-creased.The anti-inflammatory effect of Simiaoyong'an Decoction was correspondingly weakened after administration of the PPARγ inhibitor GW9662.Conclusion:Simiaoyong'an Decoction exerts an inhibitory effect on adipocyte inflammation via the PPARγ/NF-κB signaling pathway.
