Association analysis of GPX4 gene polymorphisms with Helicobacter pylori infection
Objective:To analyze the association of single nucleotide polymorphisms(SNP)in glutathione peroxidase 4(GPX4)gene with Helicobacter pylori(H.pylori)infection.Methods:A total of 506 samples were selected to detect H.pylori in-fection by enzyme-linked immunosorbent assay(ELISA)and 14C Urea breath test(UBT),a positive result of either test was deter-mined as positive;GPX4 rs4807542,rs713041,rs713041 and rs3746162 were genotyped by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method;Unconditional logistic regression was used to analyze the association of each SNP with H.pylori infection.SHEsis software was used to constructed Haplotype to analyze its association with H.pylori in-fection.The relationship between SNP-SNP interaction and H.pylori infection was analyzed by generalized multi-factor dimen-sionality reduction method.Results:GPX4 rs4807542,rs713041,rs2074451 and rs3746162 were not associated with H.pylori in-fection in codominant,dominant,recessive,and overdominant models(P>0.05);Haplotypes constructed by rs4807542,rs713041,rs2074451 and rs3746162 were not founded to be associated with H.pylori infection,and these SNPs also did not have interaction in H.pylori infection(P>0.05).Conclusion:GPX4 rs4807542,rs713041,rs2074451 and rs3746162 may not play a major role in the process of H.pylori infection in Baotou Han population.
