Experimental study on mouse model of pancreatitis constructed by ligation of pancreatic duct
Objective:To compare the pancreatic pathological characteristics of acute pancreatitis(AP)and chronic pancreati-tis(CP)models constructed by pancreatic duct ligation with those constructed solely by cerulein.Methods:A single intraperitone-al injection of cerulein(50 µg/kg/body weight)was administered 48 h after duct ligation.The mice sacrificed 3 d after ligation were designated as the AP group,while those sacrificed 21 d after ligation were designated as the CP group.The AP model in-duced by cerulein was established by continuous intraperitoneal injection of cerulein 7 times(50 μg·kg-1·h-1),mice were sacrificed 24 h after first injection.The CP model induced by cerulein was established by continuous intraperitoneal injection of cerulein on the 1st,3rd,and 6th day of the week,6 times a day(50 μg·kg-1·h-1),for a total of 6 weeks,and mice were sacrificed 5 d after last injection.H&E staining is used to detect the pathological morphology of the pancreas,immunohistochemistry staining is used to detect the expression of inflammatory mediators and fibrosis markers,and Masson staining is used to detect pancreatic fibrosis.Results:Compared to the AP model induced by cerulein,those constructed by ligation of pancreatic duct exhibited a higher patho-logical score and higher expression of inflammatory mediators IL-6 and TNF-α.Compared to the CP model induced by cerulein,those constructed by ligation of pancreatic duct exhibited more obvious loss of pancreatic acinar cells,larger proportion of fibrosis,and higher expression of fibrotic markers including α-SMA and CTGF.Conclusion:Compared to the pancreatitis model induced by cerulein,pancreatitis model constructed by ligation of pancreatic duct has severer pancreatic damage,while the CP model con-structed by it has more obvious pancreatic acinar loss and fibrosis,with lower costs in time and economy than those induced by ceru-lein.
