响应面法优化白及多糖提取工艺

目的 通过响应面法对白及多糖的水提醇沉工艺进行探究,并对最佳工艺下提取出的白及粗多糖进行分离纯化.方法 以多糖产率为指标,采用响应面分析法对白及多糖水提醇沉工艺进行优化.白及多糖采用水提醇沉法进行提取,对提取温度、提取时间、料液比和提取次数4项进行单因素考察,选取提取温度(A)、提取时间(B)和料液比(C)3个影响较大因素,以白及多糖含量提取率为响应面值W,对提取白及多糖的工艺条件进行3因素3水平响应面分析.随后依次使用DEAE-650柱和Sepharose-6B柱对白及粗多糖进行分离纯化.结果 白及多糖提取工艺影响因素:B>A>C,最佳提取条件为:提取温度68 ℃、提取时间94 min、料液比1∶14.所提取出的白及粗多糖,通过柱层析法分离纯化最终得到中性多糖组分BSP-B.结论 此提取方法操作简单,多糖含量提取率较高,适用于白及多糖提取;再通过柱层析法分离纯化粗多糖,得到组分BSP-B,为后续实验研究奠定了基础.
Optimize the Extraction Technology of Bletilla striata Polysaccharide by Response Surface Method
OBJECTIVE To investigate theprocess of water extraction and alcohol precipitation of Bletilla stria-ta crude polysaccharides by response surface method,and to separate and purify Bletilla striata crude polysaccharides extracted under the optimal process.METHODS Response surface analysis was used to optimize the process of wa-ter extraction and alcohol precipitation of white and polysaccharide with the yield of polysaccharide as index.The ex-traction temperature,extraction time,solid-liquid ratio and extraction times were investigated as single factors.The ex-traction temperature(A),extraction time(B)and solid-liquid ratio(C)were selected as three major influencing factors.With the extraction rate of polysaccharide content as response value W,the response surface analysis of three factors and three levels of extraction conditions of polysaccharide was carried out.Subsequently,DEAE-650 column and Sepharose-6B column were used to separate and purify Bletilla striata crude polysaccharides.RESULTS The influencing factors of polysaccharide extraction were as follows:B>A>C,the optimum extraction conditions were:extraction temperature 68 ℃,extraction time 94 min,solid-liquid ratio 1:14.Bletilla striata crude polysaccharides were separated and purified by column chromatography to obtain the neutral polysaccharide component BSP-B.CONCLUSION This method is easy to operate and the extraction rate of polysaccharide content is high,which is suitable for the extraction of polysaccharide.Then the crude polysaccharide was separated and purified by column chromatography to obtain the component BSP-B,which laid the foundation for the subsequent experimental study.
