不同发酵天数赤芝钩吻的毒性及镇痛作用研究

摘要
目的 比较2 批不同发酵天数钩吻的急性毒性及镇痛药效.方法 采用改良寇氏法测定发酵14 天、21 天钩吻半数致死量(LD50),并获取2 批不同发酵钩吻的最大安全给药剂量为药效学浓度.采用醋酸扭体法测定不同发酵钩吻对醋酸所致小鼠扭体反应疼痛阈值的影响,ELISA法检测各组小鼠血清中TNF-α、IL-1β和IL-6 水平.结果 发酵14、21 天的赤芝钩吻的LD50分别为10.91 g·kg-1、15.4 g·kg-1;2 批发酵钩吻对醋酸扭体镇痛模型的镇痛率分别为74%、79%,发酵钩吻显著降低血清炎症因子含量,且批次之间无显著差异.结论 2 批不同发酵天数赤芝钩吻毒性降低,并具有显著的镇痛作用,2 批次之间镇痛药效无显著差异.
Abstract
OBJECTIVE To study the toxicity and analgesic effects of Gelsemium elegans with different fermen-tation days.METHODS The modified Kou's method was used to determine the median lethal dose(LD50)of Gel-semium elegans fermented for 14 and 21 days,and the maximum safe dosage of the two batches of Gelsemium elegans was obtained.The acetic acid writhing method was used to determine the analgesic effect of fermented Gelsemium ele-gans,and the ELISA method was used to detect TNF-α,IL-1β and IL-6 levels in the serum of mice.RESULTS The LD50 of fermented Gelsemium elegans for 14 and 21 days was 10.91 and 15.4 g·kg-1,respectively;The analge-sic rates of the two batches of fermented Gelsemium elegans in the acetic acid writhing model were 74%and 79%,respectively.By comparison with the model group,two batches of fermented Gelsemium elegans reduced the content of serum inflammatory factors significantly,but there was no significant difference between the two bat-ches.CONCLUSION Gelsemium elegans showed reduced toxicity and significant analgesic effects after 14 or 21 days fermentation.
