Chemical Component,Anti-inflammatory and Antioxidant Activity in vitro of Volatile Oil from Pollen Typhae
OBJECTIVE To investigate the chemical components,anti-inflammatory and antioxidant activity of volatile oil from Pollen Typhae.METHODS The volatile oil was extracted by refluxing extraction with ether and the gas purge microinje extraction(GP-MSE).The volatile oil was analyzed by gas chromatograph-mass spectrometer(GC-MS).Area normalization method was used for quantitative analysis of the volatile oil.MTT assay was used to as-sess the cytotoxicity of volatile oil on RAW264.7.Cellular inflammation was induced by LPS treatment,the content of nitric oxide was determined by griess assay to evaluate the anti-inflammatory activity.The antioxidant activity of vola-tile oil was evaluated by scavenging capacities of DPPH free radicals,and reducing capacity of Fe3+.RESULTS Sixty-seven compounds were isolated from volatile oil of Pollen Typhae,and fifty-three compounds of them were iden-tified,which accounts for 96.89%of the total volatile oils.The main components of the volatile oil included linoleic acid,palmitic acid methyl ester,palmitic acid,linoleic acid methyl ester,oleic acid methyl ester,(Z)-9-tetradecenal and octadecanoic acid.Moreover,the content of linoleic acid was the highest(32.01%).Within the concentration range of this study,the result of griess assay showed that the volatile oil reduced the content of nitric oxide in LPS-in-duced RAW264.7 cells,with a dose-dependent manner.The result of in vivo antioxidant study showed that the vola-tile oil had no obvious scavenging capacity of DPPH free radical,as well,had no obvious reducing capacity of Fe3+.CONCLUSION The main component of volatile oil from Pollen Typhae consisting of aliphatic acid and its ester,the volatile oil had certain anti-inflammatory activity,but had not showed obvious antioxidant activity in vitro.
