紫外可见分光光度法测定牡蛎中总氨基酸的含量

目的 建立中药牡蛎中总氨基酸的含量测定方法.方法 采用紫外可见分光光度法,以茚三酮乙醇溶液和磷酸盐缓冲液为显色剂,在563nm波长下,测定牡蛎中总氨基酸的含量.结果 甘氨酸在 20.6~412 μg·mL-1 范围内线性关系良好(r = 0.9997),加样回收率为98.59%,RSD =2.95%(n =6).测得的6 批不同厂家和批号的牡蛎饮片总氨基酸含量分别为1.88mg·g-1、2.24 mg·g-1、1.76 mg·g-1、2.15 mg·g-1、2.03 mg·g-1和2.04 mg·g-1.结论 建立的测定方法简便、准确.灵敏、重复性良好,适用于牡蛎中总氨基酸含量的测定.
OBJECTIVE To establish the method for determination of the total amino acid in oyster shell.METHODS Determination of the total amino acids in oyster shell by ultraviolet visible spectrophotome-try.The chromogenic agents were ninhydrin ethanol solution and phosphate buffer.The detection wavelength was 563 nm.RESULTS It showed a good linear relationship of glycine within the range of 20.3-412μg·mL-1(r = 0.9997).The sample recovery was 98.59%,RSD =2.95%(n =6).The total amino acids in oyster shell come from 6 different manufactures and batches were 1.88 mg·g-1,2.24 mg·g-1,1.76 mg·g-1,2.15 mg·g-1,2.03 mg·g-1 and 2.04 mg·g-1 respectively.CONCLUSION The established method is simple,accuracy,sensitivity and repeatability,which was suitable for determination of the total amino acid in oyster shell.
