Optimization of the Extraction Process and Quantitative Analysis of Total Flavonoids from Schizonepetae Spica
OBJECTIVE To study the optimal extraction process of total flavonoids from Schizonepetae Spica(SS),and then to extract and quantitative analysis of total flavonoids in SS by the optimized extraction process,so as to provide scientific basis for improving the quality standard of SS.METHODS A single-factor test was carried out with the content of total flavonoids as the evaluation index,ethanol concentration,extraction time and extraction tem-perature as the investigation factors,and three optimal levels of each factor were obtained.Subsequently,a 3-factors and 3-levels L9(34)orthogonal experiment was designed,and visual analysis and variance analysis were carried out to obtain the optimal extraction process.Finally,the optimal extraction process was used to extract and determine the content of total flavonoids from 10 batches of SS.RESULTS The optimal extraction process of SS was 60%ethanol concentration,20 min extraction time and 80℃extraction temperature.The average content of total flavonoids in 10 batches SS was 5.82 mg·g-1.CONCLUSION The extraction process of total flavonoids from SS was optimized by single-factor test and orthogonal design experiments.The content of total flavonoids from 10 batches SS was deter-mined and analyzed.The limit standard of the total flavonoids content of SS was not less than 4.0 mg·g-1.
