Optimization of Extraction Technology and Antioxidant Activity of Polysac-charides from Goodyera biflora
OBJECTIVE To optimize the best extraction process of polysaccharide from Goodyera biflora(GBP)and evaluate its antioxidant activity.METHODS On the basis of single factor test results,the extraction process of GBP was optimized by orthogonal experimental design with the extraction yield of polysaccharides as the index.The antioxidant activity of the polysaccharides from Goodyera biflora were evaluated by measuring DPPH radi-cal,ABTS radical,and hydroxyl radical.RESULTS The results showed that the optimal extraction conditions of polysaccharide from Goodyera biflora were as follows:the ratio of solid to liquid was 1∶25(g·mL-1),extraction time 150 min,extraction temperature 100 ℃ and extraction repeated 3 times.Under the optimal extraction conditions,the extraction yield was 9.01%±0.51%.The results of in vitro antioxidant experiments showed that GBP had the effect of scavenging DPPH free radicals,ABTS free radicals and hydroxyl free radicals.At a polysaccharide concentration of 4 mg·mL-1,its scavenging of DPPH radicals,ABTS radicals and hydroxyl radicals was 46.42%,62.73%and 48.53%,respectively.CONCLUSION The optimized process was feasible,and the polysaccharides extracted un-der this process had certain antioxidant activity.
