五气朝阳草标准汤剂的制备及质量标准研究

为了实现五气朝阳草标准汤剂的质量控制,采用高效液相色谱法,以Waters Symmetry C18(4.6 mm× 250 mm,5.0 µm)为色谱柱,乙腈-0.1%磷酸为流动相,梯度洗脱,流速为1.0 mL/min,检测波长为360 nm,柱温为35 ℃,建立五气朝阳草标准汤剂特征图谱及其指标成分鞣花酸的含量测定方法.结果表明:15批五气朝阳草标准汤剂的平均出膏率为17.8%;特征图谱相似度为0.793~1.000,共标定13个共有峰,并指认4个成分,分别为咖啡酸(峰4)、鞣花酸(峰8)、异槲皮苷(峰9)、木犀草苷(峰10);鞣花酸从五气朝阳草到其标准汤剂的平均转移率为45.90%,说明标准汤剂的质量得到了较好的控制.所提出的五气朝阳草标准汤剂的制备工艺可行,建立的标准汤剂特征图谱及含量测定方法稳定,可为五气朝阳草配方颗粒的质量标准研究提供依据.
Study on the preparation and quality standard of Geum japonicum Thunb.var.chinense Bolle standard decoction
Inorder to realize quality control of the standard decoction of Geum japonicum Thunb.var.chinense Bolle,a method of characteristic chromatogram of Geum japonicum Thunb.var.chinense Bolle standard decoction and determination of ellagic acid was established by HPLC.The analysis was performed on Waters Symmetry C18 column(4.6 mm × 250 mm,5.0 μm)with acetonitrile-0.1%phosphoric acid for gradient elution.The flow rate was 1.0 mL/min,the detection wavelength was 360 nm and the column temperature was 35 ℃.The results show that the average extraction rate of 15 batches of Geum japonicum Thunb.var.chinense Bolle standard decoction is 17.8%;The characteristic chromatogram similarity is 0.793~1.000 with a total of 13 common characteristic peaks calibrated and four components confirmed,namely caffeic acid(peak 4),ellagic acid(peak 8),isoquercitrin(peak 9),and luteolin(peak 10);The average transfer rate of ellagic acid from Geum japonicum Thunb.var.chinense Bolle to its standard decoction is 45.90%,indicating that the quality of the standard decoction has been well controlled.The preparation process of the proposed Geum japonicum Thunb.var.chinense Bolle standard decoction is feasible,and the established characteristic chromatogram and content determination method of standard decoction are stable,which can lay a foundation for the quality standard research of the formulation granule of Geum japonicum Thunb.var.chinense Bolle standard decoction.
