分光光度法测定黄花草木樨中总黄酮含量

采用分光光度法测定了黄花草木樨中总黄酮的含量.以芦丁为对照品,NaNO2-Al (NO3)3-NaOH体系显色,在波长510 nm处对样品中的总黄酮进行含量测定.结果表明,总黄酮在10~50 μg·mL-1 (R=0.9992)范围内线性关系良好,平均回收率为99.19%(RSD=0.79%,n=9),测得样品中总黄酮平均含量为1.44%.该方法操作简便、准确,具有很好的重现性和稳定性,适用于黄花草木樨中总黄酮含量的测定.
Determination of Total Fiavonoids Content in Melilotus by Spectrophotometry
The content of total flavonoids in Melilotus was determined by spectrophotometry. Rutin was used as the standard sample and NaNO2-Al(NO3)3-NaOH was used to compose the chromogenic system. The detection wavelength was set at 510 nm. A good linear relationship was observed in the range of 10-50 μg · mL-1 ($ = 0. 9992) and the average recovery rate was 99. 19% with a relative standard deviation of 0. 79%(rt= 9). The average content of total flavonoids in Melilotus was 1. 44%. This method is simple,reliable,accurate and rapid with good reproducibility and stability. It is a suitable way for determination of total flavonoids in Melilotus.
