壳聚糖递药系统在肿瘤光动力治疗中的应用

光动力学疗法(PDT)以其良好的激发响应性和高效的肿瘤杀伤力,成为肿瘤治疗领域的研究焦点,但由于光敏剂(PS)存在溶解度低、易聚集、缺乏靶向性、治疗深度浅等缺点,阻碍了 PDT在临床应用中的进一步推广。壳聚糖(CS)作为一种天然阳离子多糖,优秀的酸碱敏感性、生物安全性、生物相容性、生物可降解性使其成为药物递送载体的理想选择。从纳米颗粒、纳米脂质体、纳米水凝胶3个方面综述了 CS及其衍生物作为光敏剂药物递送载体的应用研究进展,对CS及其衍生物递药系统在PDT治疗肿瘤中的靶向性、免疫辅助等生物学功能进行了总结,并对研究前景进行了展望,为CS与PDT联合治疗肿瘤提供了理论基础。
Application of Chitosan Drug Delivery Systems in Tumor Treatment by Photodynamic Therapy
Photodynamic therapy(PDT)has become a research hotspot in tumor treatment due to its superi-or response and high efficacy in tumor killing.However,the low solubility,propensity to aggregate,lack of tar-get specificity,and limited treatment depth of photosensitizer(PS)have hindered the further clinical application of PDT.Chitosan(CS),a natural cationic polysaccharide,has excellent acid-base sensitivity,biosafety,biocom-patibility,and biodegradability,and becomes an ideal choice for drug delivery carriers.In this paper,we review the research and application progress of CS and its derivatives as PS drug delivery carrier from three aspects:nanoparti-cles,nanoliposomes,and nanohydrogels.Moreover,we summarize the biological functions of CS and its derivatives drug delivery systems in tumor treatment by PDT,such as targetability and immune adjuvant,and put forward research prospects,aiming to provide theoretical basis for the combined application of CS and PDT in tumor treatment.
