Process Conditions for Enzyme-Catalyzed Synthesis of 2'-O-Methylguanosine
Taking 2-amino-2'-O-methyladenosine as a raw material,and adenosine deaminase(ADA)as a catalyst,we synthesized the intermediate of nucleic acid drug 2'-O-methylguanosine by an enzyme-catalyzed method.Moreover,we studied the effects of ADA dosage,reaction temperature,and pH value of buffer solution on the reaction.The results show that the content of 2'-O-methylguanosine can reach the highest of 80.00%in 44 h under the optimal conditions as follows:the ADA dosage of 150-200 U·(g 2-amino-2'-O-methylade-nosine)1,the reaction temperature of 40 ℃,and the pH value of buffer solution of 7.45.The study provides data support for the industrial production of 2'-O-methylguanosine.
