基于网络药理学和蛋白模块分析桂枝汤防治冻伤作用机制

目的 基于网络药理学和蛋白模块分析理论探究桂枝汤防治冻伤的关键靶点及关键模块,并阐述其作用机制.方法 利用TCMSP数据库筛选出满足条件(O B≥30%和DL≥0.18)的桂枝汤中药材活性化合物,通过BATMAN数据库预测所得活性成分的靶点.通过CTD、GeneCards、OMIM三大疾病数据库查找冻伤相关基因,并利用UniProt数据库对靶点进行标准化.通过St ring数据库对桂枝汤与冻伤的共有靶点进行GO和KEGG通路富集分析.借助Cytoscape3.7.0软件对桂枝汤"药材-活性化合物-靶点-通路"进行加权网络构建,获取关键靶点.再运用层次聚类凝聚算法对蛋白相互作用网络进行模块化分析,结合关键靶点筛选关键模块,并对各个模块进行富集分析.结果 在五味中药里共筛选出146种符合要求的化合物,预测得到602个桂枝汤成分的药物靶点、744个冻伤的疾病靶点,取交集后得到45个靶点.构建"活性化合物-靶点-通路"加权网络筛选出关键靶点15个,主要包括TNF、PTGS2、IL1B等,得到关键蛋白模块一、二、三和四.其中,聚类模块一主要与肿瘤坏死因子信号通路、流体剪切应力与动脉粥样硬化、花生四烯酸的新陈代谢有关,此类通路均作用于血管内皮细胞;聚类模块二主要与炎症反应的调节和凝血系统的调节有关;聚类模块三主要与氧化代谢反应及产热有关;聚类模块四主要与外界因子的刺激调节有关.结论 桂枝汤可以通过干预皮肤血管内皮细胞、炎症反应、凝血与血栓的形成、氧化代谢反应及产热等相关的生物过程发挥防治冻伤的作用.
Exploring the mechanism of Guizhi Tang in preventing and treating frostbite based on network pharma-cology and protein module analysis
Objective To explore the key targets and modules of Guizhi Tang in preventing and treating frostbite based on network pharmacology and protein module analysis theory,and elaborate its mechanism of action.Methods The active compounds of traditional Chinese medicinal herbs in Guizhi Tang that meet the conditions(OB≥30%and DL≥0.18)were screened using the TCMSP da-tabase.The targets of the obtained active ingredients were predicted through the BATMAN database.Frostbite-related genes were searched through the CTD,GeneCards,and OMIM disease databases,and the targets were standardized using the UniProt database.GO and KEGG pathway enrichment a-nalysis was performed on the common targets of Guizhi Tang and frostbite using the String database.Cytoscape 3.7.0 software was used to construct a weighted network of"herb-active compound-target-pathway"of Guizhi Tang to obtain key targets.Hierarchical clustering and agglomerative algorithms were then used to perform modular analysis on the protein-protein interaction network,and key mod-ules were screened based on the key targets.Enrichment analysis was performed on each module.Re-sults A total of 146 compounds meeting the requirements were screened from the five traditional Chinese medicinal herbs.602 drug targets of Guizhi Tang components and 744 disease targets of frostbite were predicted,and 45 targets were obtained after taking the intersection.Fifteen key tar-gets were screened out from the constructed"active compound-target-pathway"weighted network,mainly including TNF,PTGS2,IL1B,etc.Four key protein modules were obtained,namely cluste-ring modules 1,2,3,and 4.Among them,clustering module 1 is mainly related to tumor necrosis factor signaling pathways,fluid shear stress and atherosclerosis,and arachidonic acid metabolism,all of which act on vascular endothelial cells.Clustering module 2 is mainly related to the regulation of inflammatory responses and the coagulation system.Clustering module 3 is mainly related to oxi-dative metabolic reactions and heat production.Clustering module 4 is mainly related to the regula-tion of stimulation by external factors.Conclusion Guizhi Tang can play a role in preventing and treating frostbite by intervening in biological processes related to skin vascular endothelial cells,in-flammatory responses,coagulation and thrombus formation,oxidative metabolic reactions,and heat production.
