枯草芽孢杆菌B1514菌株发酵条件的优化与片剂制作研究

为提高枯草芽孢杆菌B1514菌株的发酵产量,降低生产成本,方便田间应用,本研究通过正交试验设计和单因素试验筛选的方法对其进行了液体发酵条件和吸附载体配比的优化,并通过筛选湿法制粒的最优筛目和润滑剂硬脂酸镁的用量,对其片剂制作进行了初步尝试.结果表明:适合该菌株的最佳培养基配方为葡萄糖1.5%,淀粉0.5%,玉米粉3%,硫酸锰0.2%,硫酸铵1.5%,硫酸镁0.02%,磷酸二氢钾0.03%,氯化钙0.1%;最佳发酵条件为:接种量5%,装液量100 mL/500 mL,初始pH 7.0~7.5,发酵温度32℃.发酵条件优化后活菌含量增至1.17×1010 cfu/mL,产量提高115%.以玉米粉、玉米秸秆粉、麦麸(质量比1∶6∶2)为吸附载体,20目筛制粒,1%的硬脂酸镁为润滑剂,可制得片形完整、硬度适宜,符合标准的片剂.
Optimization of fermentation conditions and the preperation of tablets for Bacillus subtilis strain B1514
In order to improve the production of Bacillus subtilis strain B1514,reduce production cost and make it convenient for application,single factor and orthogonal experimental designs were carried out to optimize the fermentation conditions and the proportion of carriers,and the preperation conditions of tablets for B1514 were optimized by screening the mesh for wet granulation and the dosage of lubricant magnesium stearate.The results showed that the optimum fermentation medium components for B.subtilis strain B1514 were glucose 1.5%,amylon0.5%,corn flour 3%,MnSO4 0.2%,(NH4)2SO4 1.5%,MgSO4 0.02%,KH2PO4 0.03 % and CaCl2 0.1%,respectively; the optimum fermentation conditions were that putting 5% initial inoculums into 100 mL liquid culture in a 500 mL flask,with the initial pH 7.0-7.5 and keeping the fermentation temperature at 32 ℃.Under the optimum conditions,the number of live bacterium increased to 1.17 × 1010 cfu/mL,and the yield of fermentation increased by 115%.Using corn flour,corn stalk powder,and wheat bran(mass ratio 1 ∶ 6 ∶ 2) as carriers,20 oculus screen for wet granulation and 1% magnesium stearate as lubricant,can get tablets of B1514 up to standard with intact shape and suitable rigidity.
