本研究以'中薯 18 号'为试材,分别在短日照(8 h light/16 h dark)和长日照(16 h light/8 h dark)条件下进行培养,定期观察匍匐茎顶端的生长变化,取不同光照条件下匍匐茎顶端弯钩期、亚顶端初始膨大期及亚顶端膨大期的叶片和匍匐茎亚顶端部位进行转录组测序和分析发现长日照条件下匍匐茎亚末端初始膨大比短日照条件下显著延迟,昼夜节律通路在各个阶段都有富集,与光周期调控块茎初始形成密切相关.22 个Dof、ERF、MYB等家族的转录因子在匍匐茎亚顶端初始膨大期显著上调表达的模块,可能参与调控马铃薯块茎初始形成.本研究为深入解析光周期调控马铃薯块茎初始形成的遗传调控机制提供了重要参考.
Transcriptome analysis of tuber initiation of Solanum tuberosum regulated by photoperiod
We grew the photoperiod sensitive material'Zhongshu 18'under short day(8 h light/16 h dark)and long day(16 h light/8 h dark)respectively in this study.The growth changes of the stolon apex were observed regularly.Transcriptome sequencing and analysis were performed on the leaves and apex of the stolons when the stolon hooked,and the stolon below the apex initiated swelling and displayed swelling under different photoperiods.The results showed that the stolon below the apex initiated swelling significantly late under long day compared to that under short day.The circadian rhythm pathway was enriched at all stages and closely related to the photoperiod regulation of tuber initiation.22 transcription factors of Dof,ERF,MYB and other families were significantly up-regulated during the initiated swelling of stolon below the apex,suggesting their involvement in regulation of potato tuber initiation.This study provideed an important reference for in-depth analysis of the genetic regulation of photoperiod on the potato tuber initiation.
