STAT3的多态性在HPV16所致宫颈癌发生中的作用

目的:研究人乳头状瘤病毒16(HPV16)所致宫颈癌发生过程中转录活化子3(STAT3)多态性的作用。方法:回顾性选取2019 年至2023 年入院的34 例宫颈癌患者为观察对象,将其设为A组,选取同一时期入院的54 例宫颈上皮内瘤变患者将其设为B 组,同时选取同一时期入院的 80 例慢性宫颈炎患者将其设为C组。HPV16 E6 蛋白测定方式主要为免疫组化、聚合酶链反应(PCR)-反向点杂交法,STAT3 测定方式主要为PCR法,STAT3 基因C1697G多态性测定方式主要为PCR-制片段长度多态性(RFLP),比较并分析三组患者E6 蛋白、STAT3 蛋白阳性表达率与 STAT3 基因多态性表达情况。结果:与B、C组相比,A组患者E6、STAT3 蛋白阳性表达率明显更高,差异有统计学意义(P<0。0167);相比C组患者,B组患者E6 蛋白、STAT3 蛋白阳性表达率明显更高,差异有统计学意义(P<0。0167)。相比C组患者,A、B 组患者 G/G 型构成比明显更低,C/C 型构成比明显更高,差异有统计学意义(P<0。0167);相比 B 组患者,A 组患者 G/G 型构成比明显更低,C/C 型构成比明显更高,差异有统计学意义(P<0。0167);三组患者G/C型构成比差异无统计学意义(P>0。0167)。结论:HPV16 所致宫颈癌发生及发展过程中STAT3 及其多态性可能有重要作用。
The Role of STAT3 Polymorphism in the Development of Cervical Cancer Caused by HPV16
Objective:To investigate the role of signal transducer and activator of transcription 3(STAT3)polymorphism in the development of cervical cancer caused by human papillomavirus 16(HPV16).Methods:A retrospective study was conducted on 34 patients with cervical cancer who were admitted to the hospital from 2019 to 2023.They were assigned to Group A.Another 54 patients with cervical intraepithelial neoplasia and 80 patients with chronic cervicitis admitted to the hospital during the same period were assigned to Groups B and C,respectively.The expression of HPV16 E6 protein was detected by immunohistochemistry and polymerase chain reaction(PCR)-reverse dot blot hybridization.The expression of STAT3 protein was detected by PCR,and the polymorphism of STAT3 gene C1697G was detected by PCR-restriction fragment length polymorphism(RFLP).The expression rates of E6 and STAT3 proteins and the genotypes of STAT3 gene polymorphism in the three groups were compared and analyzed.Results:The positive expression rates of E6 and STAT3 proteins in Group A were significantly higher than those in Groups B and C(P<0.0167).The positive expression rates of E6 and STAT3 proteins in Group B were significantly higher than those in Group C(P<0.0167).The frequency of the G/G genotype in Groups A and B was significantly lower than that in Group C,and the frequency of the C/C genotype was significantly higher than that in Group C(P<0.0167).The frequency of the G/G genotype in Group A was significantly lower than that in Group B,and the frequen-cy of the C/C genotype was significantly higher than that in Group B(P<0.0167).There was no significant difference in the frequency of the G/C genotype among the three groups(P>0.0167).Conclusion:STAT3 and its polymorphism may play an important role in the development and progression of cervical cancer caused by HPV16.
