LncRNA H19靶向调控miR-19b-3p/SOX9通路对脂肪间充质干细胞向成骨细胞分化的影响

目的:分析长链非编码核糖核酸(LncRNA)H19靶向调控miR-19b-3p/性别决定区Y框转录因子9(SOX9)通路对脂肪间充质干细胞(ADMSC)向成骨细胞分化的影响。方法:体外培养人ADMSC细胞并鉴定,将ADMSC细胞分为对照组(常规培养)、H19组(转染LncRNA H19慢病毒过表达载体)、LncRNA-NC组(转染慢病毒空载体)、miR-19b-3p组(共转染LncRNA H19慢病毒过表达载体和miR-19b-3p模拟物慢病毒载体)、miR-NC组(共转染LncRNA H19慢病毒过表达载体和 miR-NC慢病毒载体),检测各组LncRNA H19、miR-19b-3p、SOX9及含锌指的成骨细胞特异性转录因子(Osx)、骨钙素(OCN)和RUN相关转录因子2(RUNX2)表达。观察各组ALP染色和茜素红染色情况,比较细胞增殖活力、ALP活性,分析各组SOX9、BMP-2、OPN 蛋白表达,验证LncRNA H19和 miR-19b-3p的靶向关系。结果:与对照组、LncRNA-NC 组比,H19 组 LncRNA H19 和 SOX9、Osx、OCN、RUNX2 的 mR-NA表达及SOX9、BMP-2、OPN蛋白表达、ALP活性、矿化结节形成量增加,miR-19b-3p表达降低(P<0。05),与 H19 组、miR-NC 组比,miR-19b-3p 组 LncRNA H19 和 SOX9、Osx、OCN、RUNX2 的 mRNA 表达及SOX9、BMP-2、OPN蛋白表达、ALP活性、矿化结节形成量降低,miR-19b-3p表达增加(P<0。05)。各组细胞24h、48h、72h的吸光度比较,差异无统计学意义(P>0。05)。LncRNA H19和 miR-19b-3p间存在靶向关系。结论:LncRNA H19能负向调控miR-19b-3p,上调SOX9,有助于人ADMSC细胞向成骨细胞分化。
LncRNA H19 Targets miR-19b-3p/SOX9 Pathway to Promote Osteogenic Differentiation of Adipose-Derived Mesenchymal Stem Cells
Objective:To investigate the effects of LncRNA H19 targeting miR-19b-3p/SOX9 pathway on the osteogenic differentiation of adipose-derived mesenchymal stem cells(ADMSCs).Methods:Human ADMSCs were cultured and identified in vitro.The cells were divided into five groups:control group(regular culture),H19 group(transfected with LncRNA H19 lentivirus overexpression vector),LncRNA-NC group(transfected with lentivirus empty vector),miR-19b-3p group(co-transfected with LncRNA H19 lentivirus overexpression vector and miR-19b-3p mimic lentivirus vector),and miR-NC group(co-transfected with LncRNA H19 lentivirus overexpression vector and miR-NC lentivirus vector).The expression of LncRNA H19,miR-19b-3p,SOX9,zinc finger osteoblast-specific transcription factor(Osx),osteocalcin(OCN),and RUNX2 was detected in each group.ALP staining and Alizarin Red staining were observed in each group,cell proliferation activity and ALP activity were compared,and the expression of SOX9,BMP-2,and OPN proteins in each group was analyzed to verify the targeted relationship between LncRNA H19 and miR-19b-3p.Results:Compared with the control group and LncRNA-NC group,the expression of LncRNA H19 and SOX9,Osx,OCN,and RUNX2 mRNA,as well as the expression of SOX9,BMP-2,and OPN proteins,ALP activity,and the amount of mineralized nodule formation were increased in the H19 group,while the ex-pression of miR-19b-3p was decreased(P<0.05).Compared with the H19 group and miR-NC group,the expression of LncRNA H19 and SOX9,Osx,OCN,and RUNX2 mRNA,as well as the expression of SOX9,BMP-2,and OPN proteins,ALP activity,and the amount of mineralized nodule formation were decreased in the miR-19b-3p group,while the expression of miR-19b-3p was increased(P<0.05).There was no statis-tical difference in the absorbance of each group at 24h,48h,and 72h(P>0.05).There was a targeted rela-tionship between LncRNA H19 and miR-19b-3p.Conclusion:LncRNA H19 can negatively regulate miR-19b-3p,upregulate SOX9,and contribute to the differentiation of human ADMSC cells into osteoblasts.Ln-cRNA H19 can negatively regulate miR-19b-3p,upregulate SOX9,and promote the osteogenic differentia-tion of human ADMSCs.
