Correlation Analysis of the Expression Levels of Transmembrane Protein Klotho and Fibronectin in Keloids and Their Relationship with Postoperative Recurrence
Objective:To investigate the correlation between Klotho protein binding to fibronectin Fn and postoperative recurrence in keloids.Methods:A total of 43 patients with ear keloids and 37 general cos-metic seekers who visited our department from April 2020 to April 2023 were selected.The subjects were di-vided into a study group(keloid tissues and serum)and a control group(normal tissues and serum).Immu-nohistochemistry and enzyme-linked immunosorbent assay(ELISA)were used to detect and compare the ex-pression of Klotho protein and the epithelial-mesenchymal transition marker fibronectin FN in tissue and serum samples from both groups.Keloid patients were followed up at 1 month,3 months,6 months,and 12 months postoperatively to record recurrence,which divided them into recurrence and non-recurrence groups.Statisti-cal methods were used to analyze the correlation between Klotho expression and fibronectin FN expression and postoperative recurrence in keloids.Results:Immunohistochemistry results showed that the positive rate of Klotho in the study group was lower than in the control group(P<0.05),at 37.20%.The positive rate of FN in the study group was 72.10%,higher than in the control group(P<0.05).The positive rate of Klotho in the recurrence group was 21.40%,lower than in the non-recurrence group(P<0.01),and the positive rate of FN in the recurrence group was 92.90%,higher than in the non-recurrence group(P<0.05).There was a statistically significant correlation between the levels of Klotho and FN proteins(r=-0.307,P<0.05).Elisa results showed that the expression level of Klotho in the study group was 65.99(55.05,79.06)ng/mL,lower than in the control group(P<0.05),and the expression level of FN in the study group was 83.56(62.67,91.85)ng/mL,higher than in the control group(P<0.01).Logrank tests indicated that negative Klotho expression and positive FN protein expression were associated with postoperative recurrence(both P<0.05).Conclusion:The combination of Klotho protein and the epithelial-mesenchymal transition marker FN can be used as an important marker to predict postoperative recurrence of keloids.
