Experimental Study on the Effect of Silencing HIF-1α on the Growth of Laryngeal Cancer Cells by Regulating SLC7A11
Objective:To explore the specific effect of silencing HIF-1α on SLC7A11 in laryngeal canc-er,and to find new tumor markers and potential therapeutic targets for clinical diagnosis of laryngeal cancer.Methods:UMSCC5 laryngeal cancer cells were cultured and transfected with HIF-1α-siRNA.Western blot was used to screen the best transfection efficiency.CCK-8 was used to detect the proliferation activity of the transfected cells.Flow cytometry was used to detect the apoptosis of each group.Transwell method was used to detect cell invasion and migration.Western blot was used to detect the expression of HIF-1α and SLC7A11.Results:The expression of HIF-1α was successfully silenced,and the transfection efficiency of HIF-1α-siR-NA1 group was the highest and the most stable(P<0.05).Silencing HIF-1α can significantly inhibit the proliferation of human laryngeal carcinoma UMSCC5 cells(P<0.05),promote apoptosis(P<0.05),and inhibit the invasion and migration of laryngeal carcinoma cells(P<0.05).Silencing HIF-1α could signifi-cantly down-regulate the expression of SLC7A11 protein(P<0.05).Conclusion:Silencing HIF-1α can inhibit the proliferation,invasion,migration and apoptosis of human laryngeal carcinoma UMSCC5 cells,and inhibit the expression of SLC7A11.It provides new ideas and theoretical basis for clinical molecular targeted therapy of laryngeal cancer based on HIF-1α in the future.
