Study on the Regulation of Growth and Metastasis of Esophageal Squamous Cell Carcinoma Cells by LncRNA UCA1
Objective:To investigate the effects of LncRNA UCA1 on the in vivo and in vitro growth and metastasis of esophageal squamous cell carcinoma cells.Methods:qRT-PCR was used to detect the expres-sion of LncRNA UCA1 in human esophageal epithelial cells(HEEC),human esophageal squamous cell carci-noma cells(EC109,EC9706,KYSE150,and KYSE450).EC109 and EC9706 cells were divided into si-NC group and si-UCA1 group.CCK8 and flow cytometry were used to detect cell proliferation and cell cycle,Tr-answell method was used to detect cell migration and invasion.Western blot was used to detect the relative ex-pression levels of E-cadherin,Vimentin,and MMP-9 in cells.1×106 EC109 cells of si-NC and si-UCA1 groups were inoculated into the left axilla of BALB/c-A nude mice,after 4 weeks of feeding,the tumor vol-ume and weight of the mice were observed and recorded.Results:The expression of LncRNA UCA1 in EC109,EC9706,KYSE150,and KYSE450 was significantly higher than that in HEEC cells(P<0.001).Compared with the si-NC group,the proliferation,migration,and invasion abilities of EC109 and EC9706 cells in the si-UCA1 group were significantly reduced(P<0.05),cell cycle was significantly arrested(P<0.05),the expression of E-cadherin was significantly upregulated(P<0.001),and the expression of Vimentin and MMP-9 were significantly downregulated(P<0.001).The tumor volume and weight of mice in the si-UCA1 group were significantly lower than those in the si-NC group(P<0.001).Conclusion:Inhibiting Ln-cRNA UCA1 can inhibit the proliferation,migration,and invasion ability of esophageal squamous cell carcino-ma cells,as well as their growth in mice.
