Effect of external application of Breast Pain Healing Film on rats with mammary gland hyperplasia
Objective To observe the effect of external application of Breast Pain Healing Film(BPHF)on rats with mammary gland hyperplasia.Methods A total of 55 rats were randomly divided into control group,model group,tamoxifen ointment group,SanjieRupi Plaster(SJRPP)group and BPHF group,with 11 rats per group.Rats in the latter four groups were subjected to the modeling of mammary gland hyperplasia.After successful modeling,rats in the control group and model group were externally applied with 0.1 g blank vehicle per rat on the second pair of mammary glands.Those in the tamoxifen ointment group,SJRPP group and BPHF group were externally applied with 0.1 g tamoxifen ointment,0.4 g SJRPP(equivalent to 0.1g of the original SanjieRupi Patch)and 0.8 g BPHF(equivalent to 0.8 g of crude drug)per rat on the second pair of mammary glands,respectively.Changes in the nipple appearance of rats were observed.Spleen index and thymus index were recorded.Se-rum estradiol(E2),progesterone(P),prolactin(PRL),malon-dialdehyde(MDA)and superoxide dismutase(SOD)levels were measured.Pathological morphology of the breast tissue was visualized,and expressions of estrogen receptor α(ERα),ERβ and progesterone receptor(PR)in the breast tissue were detected.Results Compared with those of the control group,the nipple di-ameter and height of the model group significantly increased(P<0.05).Compared with those of the model group,the nipple di-ameter and height of the tamoxifen ointment group,SJRPP group and BPHF group significantly decreased(P<0.05),which were comparable among the latter three groups(P>0.05).Compared with those of the control group,the spleen index and thy-mus index of the model group significantly decreased(P<0.05).Compared with those of the model group,the spleen index and thymus index of the tamoxifen ointment group,SJRPP group and BPHF group significantly increased(P<0.05),which were comparable among the latter three groups(P>0.05).Compared with those of the control group,rats in the model group presen-ted significantly higher E2 and PRL,and lower P(both P<0.05).Compared with those of model group,rats in the tamoxifen ointment group,SJRPP group and BPHF group presented significantly lower E2 and PRL,and higher P(both P<0.05).P level was significantly higher in SJRPP group than that of tamoxifen ointment group(P<0.05).Compared with those of the control group,rats in the model group presented significantly higher MDA and lower SOD(both P<0.05).Compared with those of model group,rats in the tamoxifen ointment group,SJRPP group and BPHF group presented significantly lower MDA and higher SOD(both P<0.05).Compared with those of tamoxifen ointment group,rats in the SJRPP group and BPHF group presented significantly lower MDA and higher SOD(both P<0.05).Compared with those of the control group,rats in the model group presented significantly higher ERα,ERβ and PR in the mammary gland tissue(P<0.05).Compared with those of model group,rats in the tamoxifen ointment group,SJRPP group and BPHF group presented significantly lower ERα,ERβ and PR in the mammary gland tissue(P<0.05),which were comparable among the latter three groups(P>0.05).Conclusion BPHF can improve the hyperplasia of mammary gland tissue in rats,presenting the biological effect on regulating serum sex hormone,free radical metabolism and estrogen receptors(ERα,ERβ)in mammary gland tissue,and weakening the secretion of E2.
