Effect of Zhenwu Decoction plus geosaurus combined with cisplatin on the levels of interleukin-7 and vascular endothe-lial growth factor of mice with malignant pleural effusion
Objective To investigate the effect of combined administration of Zhenwu Decoction plus geosaurus and cis-platin on interleukin-7(IL-7)and vascular endothelial growth factor(VEGF)of mice with malignant pleural effusion(MPE).Methods A MPE model was established in C57BL/6 mice with Lewis lung carcinoma(LLC).Forty 8-week-old female C57BL/6 mice were habituated for 1 week,and they were randomly assigned into blank control group,model group,cisplatin group,Zhenwu Decoction plus geosaurus group and Zhenwu Decoction plus geosaurus+cisplatin group,with 8 mice in each group.In addition to mice in the blank control group,32 mice in the remaining groups were used to establish a MPE model using LLC cells.Mice in the blank control and model group were given 0.9%sodium chloride injections once a day for 10 days.Three days after successful modeling,mice in the cispl-atin group were intraperitoneally injected with diluted cisplatin in-jection at6 mg·kg-1·d-1,once every3 days for3 days.Mice in the Zhenwu Decoction plus geosaurus group were given 33.78 g·kg-1·d-1 crude drug solution once a day for 10 days.Mice in the Zhenwu Decoction plus geosaurus+cisplatin group were intraperitoneally injected with6 mg·kg-1·d-1 cisplatin injection,once every 3 days,and intragastatically adminis-trated with33.78 g·kg-1·d-1 crude drug solution once a day for 10 days.Mice were routinely anesthetized to expose the chest,and0.1 mL of pleural fluid was extracted with a syringe.The volume of pleural fluid was recorded.Serum levels of IL-7 and VEGF were detected by enzyme-linked immunosorbent assay(ELISA).Results Compared with the blank control group,the remaining groups had significantly increased chest water volume(P<0.05).Compared with the model control group,the drug treatment groups presented significantly decreased chest water volume(P<0.05),Zhenwu Decoction plus geosaurus+cisplatin group was superior to Zhenwu Decoction plus geosaurus group and cisplatin group for chest water volume(all P<0.05).Compared with the model blank group,the remaining groups had significantly increased VEGF(P<0.05).Compared with the model control group,the drug treatment groups presented significantly decreased VEGF(P<0.05),Zhenwu Decoction plus geosaurus+cispla-tin group had significantly decreased VEGF compared to Zhenwu Decoction plus geosaurus group and cisplatin group(all P<0.05).Compared with the blank control group,the remaining groups had significantly decreased IL-7(P<0.05).Compared with the model control group,the Zhenwu Decoction plus geosaurus group and Zhenwu Decoction plus geosaurus+cisplatin groups presented significantly increased IL-7(P<0.05),while cisplatin group presented significantly decreased IL-7(P<0.05),the Zhenwu Decoction plus geosaurus+cisplatin group and Zhenwu Decoction plus geosaurus group had significantly increased IL-7 relative to cisplatin group(P<0.05).Zhenwu Decoction plus geosaurus group had significantly increased IL-7 relative to Zhenwu Decoction plus geosaurus+cisplatin group(P<0.05).Conclusion Zhenwu Decoction plus geosaurus combined with cisplatin has a good effect on MPE in mice,which can decrease chest water volume,it may be related to the downregulation of VEGF and upregulation of IL-7 expression.In addition,which can reverse cisplatin-induced IL-7 downregulation.
