Study on the protective effect and material basis of Hugan Buzure Granules on chronic renal insufficiency mice
Objective To analyze the chemical composition of Hugan Buzure Granules(HBG)and explore its therapeutic effect on chronic renal insufficiency(CRF)mice.Methods Using the CRF mouse model induced by adenine,the levels of creatinine(Cr)and blood urea nitrogen(BUN)in the serum of mice were detected;HE staining and MASSON staining were used to detect the proliferation and damage of kidney tissue;using the chromatographic and mass spectrometric data of some reference substances,the fragmentation rules were deduced from the primary and secondary fragment ions to complete the qualitative analysis of the compounds.Results Compared with model group,HBG high dose group had the best therapeutic effect.After HBG treatment,the contents of Cr and BUN in serum of mice were significantly decreased;HE staining results showed that the histological characteristics of renal injury in HBG group were significantly improved,vacuoles were reduced,and renal tubule dilation was reduced.MASSON staining results showed that the histological characteristics of renal injury in HBG group were significantly improved,vacuoles were reduced,and fibroproliferation was reduced.A total of 83 compounds were resolved from HBG methanol extract using MassLnx V4.1 software and reference material,the most of which were flavonoids,and also contained fatty acid compounds,phenylpropanoid compounds and other types of compounds.Conclusion HBG can alleviate the renal tissue damage of CRF mice,improve the renal function and tissue fibrosis of mice,and has a certain protective effect on CRF mice.
