Improvement of Smilax china L.on croton oil-induced hemorrhoids rat model through NLRP3 pathway
Objective To investigate the effect of improvment and its mechanism of Smilax china L.on croton oil-induced hemorrhoid rats through NLRP3 pathway.Methods Forty SD rats of 6-week-old SPF grade,half male and female,were selected and randomly divided into control group,model group,low-dose Smilax china L.group,high-dose Smilax china L.group and diosmin tablet group,8 in each group.After modeling,the low-dose Smilax china L.group,high-dose Smilax china L.group anddiosmin tablet group were given 1.58 g/kg,6.3 g/kg,and 0.122 g/kg by gavage respectively,once a day for 1 week.Twenty-four hours after the last intragastric administration,the rectal tissue and serum of rats were taken.Hematoxylin eosin(HE)staining was used to detect the pathological changes of rectal tissue;test kits were used to detect the content of lactate dehydrogenase(LDH),interleukin-1β(IL-1β),IL-18,IL-10 and IL-13 in serum;the expressions of NOD-like receptor thermal protein domain associated protein 3(NLRP3),cleaved cysteinyl aspartate specific proteinase-1(Cleaved Caspase-1)and gasdermin D(GSDMD)proteins in rectal tissues were detected by Western Blot.Results Compared with control group,the rectal tissue pathological damage of model group was aggravated,the expressions of NLRP3,Cleaved Caspase-1,GSDMD,LDH,IL-1β and IL-18 were significantly increased(P<0.05),while the contents of IL-10 and IL-13 decreased significantly(P<0.05).Compared with model group,the rectal tissue pathological damage of the low-dose Smilax china L.group was alleviated,the expressions of IL-18,NLRP3 and GSDMD were significantly decreased(P<0.05),there was no significant difference in Cleaved Caspase-1,LDH,IL-1β,IL-10 and IL-13 content(P>0.05).Compared with model group,the rectal tissue pathological damage of the high-dose Smilax china L.group was alleviated,and the expressions of IL-1β,IL-18,NLRP3,Cleaved Caspase-1 and GSDMD were sig-nificantly decreased(P<0.05),there were no significant differences in Cleaved Caspase-1,LDH,IL-1β,IL-10 and IL-13 contents(P>0.05).Conclusion Smilax china L.has a good therapeutic effect on hemorrhoid rats,and its specific mechanism may be related to the regulation of NLRP3 pathway.
