Impact of galangin on vascular endothelial cell injury induced by high glucose by regulating CCL2-CCR2 signal axis
Objective To investigate the impact of galangin on the injury of vascular endothelial cells(HUVEC)induced by high glucose and the regulation mechanism on CCL2-CCR2 signal axis.Methods HUVEC was separated into control group,high glucose group,galangin low and high concentration group,galangin high concentration+pcDNA-NC,and galangin high concentration+pcDNA-CCL2 group.The proliferative activity of cells was detected by CCK-8;cell apoptosis was detected by flow cytometry;the level of ROS of cells was detected by DCFH-DA;the levels of interluskin-6(IL-6),tumor necrosis factor-α(TNF-α),chemokine ligand 2C(CCL2),malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)was detected by ELISA;the expression of Bax and Bcl-2 proteins of cells was detected by Western Blot;the levels of CCL2 and CCR2 mRNA of cells was detected by qRT-PCR.Results Compared with control group,the cell survival rate,SOD and GSH-PX activities,and expression of Bcl-2 in high glucose group decreased,the cell apoptosis rate,ROS production,levels of MDA,IL-6,TNF-α and CCL2,expression of Bax,levels of CCL2 and CCR2 mRNA increased(P<0.05);compared with high glucose group,the cell survival rate,SOD and GSH-PX activities,and expression of Bcl-2 in high glucose group increased,the cell apoptosis rate,ROS production,levels of MDA,IL-6,TNF-α,and CCL2,expression of Bax,levels of CCL2 and CCR2 mRNA decreased(P<0.05);subsequently,the cells transfected with CCL2 overexpression plasmid were resupplied,the results showed that CCL2 overexpression reversed the alleviating effect of galangin on high glucose-induced HUVEC injury(P<0.05).Conclusion Galangin can alleviate the injury of vascular endothelial cells induced by high glucose,and its mechanism may be related to the inhibition of CCL2-CCR2 signaling pathway.
