Extraction and characterization of Codonopsis pilosulae polysaccharide and its dispersion on silver nanoparticle
Objective To isolate polysaccharide from Banqiao Codonopsispilosula and characterize its structure.The polysaccharide was used to disperse and stabilize nanosilver(AgNPs),and its dispersion mechanism was to be detected.Methods Codonopsis pilosulae polysaccharide was extracted by water extraction and alcohol precipitation method,and its molecular weight distribution and monosaccharide composition was analyzed.Using an in-situ reduction approach,polysaccharide nano-silver complexes were prepared.The dispersion mechanism of BD-Ag was analyzed by X-ray diffractometer(XRD),Fourier infrared(FT-IR),and X-ray electron spectrometer(XPS).Results The yield of Codonopsis pilosula polysaccharide was 2.48%,and the molecular weights Mp,Mw and Mn were 4036,4543 and 3351 Da respectively.The Codonopsis pilosula polysaccharide was consisted of rhamnose,mannose,glucose,galactose and arabinose.When the prepared AgNPs had a zeta potential value of-14.73 mV,the optimal condition of polysaccharide AgNPs complex was 0.1 mg/mL for polysaccharide con-centration and 6 mM for AgNO3 concentration.Conclusion The Codonopsispilosulae polysaccharides can stabilize and disperse the AgNPs by forming Ag-O bonds with AgNPs.
