摘要

通过HPLC指纹图谱比较皂角子与滇皂角子的差异,并结合化学计量学方法为皂角子药材的鉴别提供依据.方法 选用Diasonsil C18(250 mm×4.6 mm,5 μm)色谱柱,以0.1%磷酸溶液-乙腈为流动相,梯度洗脱;流速为1.0 mL/min;柱温为25℃;检测波长为210 nm.采用"中药色谱指纹图谱相似度评价系统"软件(2012A版)进行数据处理分析,计算皂角子及滇皂角子的相似度.结果 建立的皂角子指纹图谱共确定了 11个共有峰;滇皂角子整体的相似度较比皂角子样品低,在0.783~0.842之间,而皂角子样品中除亳州市外,其余产地的相似度均在0.898以上.聚类分析结果将13批皂角子和3批滇皂角子样品聚为4类,主成分分析结果与聚类分析基本一致.结论 该方法能有效区分皂角子与滇皂角子,可为控制皂角子药材的质量提供参考.

Abstract

Objective To compare the differences between the Gleditsiae sinensis Semen and Gleditsiae japonica var.Delavayi Semen by HPLC fingerprint,so as to provide a basis for the identification of the Gleditsiae sinensis Semen by combining chemical chemometrics methods.Methods An Diasonsil C18(250 mm×4.6 mm,5 μm)chromatographic column was used,the mobile phase was 0.1%phosphoric acid solution and acetonitrile with gradient elution,the flow rate was 1.0 mL/min,column temperature was 25 degrees centigrade,and detection wavelength was 210 nm.The similarity evaluation software(2012A Version)of TCM chromatographic fingerprint prepared by pharmacopoeia committee was used for data processing,and the similarity between the extracts of the Gleditsiae sinensis Semen and Gleditsiae japonica var.Delavayi Semen was calculated by the angle cosine method.Results A total of 11 common peaks were calibrated in the established fingerprint.The similarity of the Gleditsiae japonica var.Delavayi Semen as a whole was lower than that of the Gleditsiae sinensis Semen samples,ranging from 0.783 to 0.842,but the similarity of the samples of the Gleditsiae sinensis Semen samples was above 0.898 except for Bozhou.The results of cluster analysis showed that 13 batches of the Gleditsiae sinensis Semen samples and 3 batches of the Gleditsiae japonica var.Delavayi Semen samples were grouped into 4 categories,and the results of principal component analysis were basically consistent with those of cluster analysis.Conclusion This method can effectively distinguish the Gleditsiae sinensis Semen and Gleditsiae japonica var.Delavayi Semen,which provides a technical guarantee for controling the quality of the Gleditsiae sinensis Semen.
