虫草保肾颗粒对肾性骨病大鼠的改善作用及Klotho/FGF-23通路的影响

目的 观察虫草保肾颗粒(CCBSG)对肾性骨病(ROD)大鼠的改善作用及Klotho/成纤维细胞生长因子-23(FGF-23)通路的影响.方法 将72只大鼠随机分为对照组、ROD组、CCBSG低剂量组、CCBSG中剂量组、CCBSG高剂量组、骨化三醇组,每组12只.除对照组外,其它组大鼠均需通过5/6肾切除联合高磷饲养的方式构建ROD大鼠模型,建模成功后,进行给药处理,给药1次/d,持续12周.自动生化分析仪检测大鼠血清中尿素氮(BUN)、血肌酐(Scr)、钙(Ca2+)、磷(P3+)水平;ELISA法检测大鼠血清中全段甲状旁腺素(iPTH)水平;双能X射线检测大鼠股骨骨密度;HE染色评估股骨组织病理变化;Western Blot检测Klotho、FGF-23蛋白表达.结果 与对照组比较,ROD组大鼠股骨病理损伤严重,血清中BUN、Scr、P3+、iPTH水平及股骨组织中FGF-23蛋白表达升高,Ca2+水平、骨密度及股骨组织中Klotho蛋白表达下调(P<0.05);与ROD组比较,CCBSG低剂量组、CCBSG中剂量组、CCBSG高剂量组、骨化三醇组大鼠股骨病理损伤减轻,血清中BUN、Scr、P3+、iPTH水平及股骨组织中FGF-23蛋白表达降低,Ca2+水平、骨密度及股骨组织中Klotho蛋白表达升高(P<0.05).结论 CCBSG改善大鼠ROD可能与调控Klotho/FGF-23通路有关.
Improving effect of Chongcao Baoshen Granules on renal osteodystrophy rats and its impact on the Klotho/FGF-23 pathway
Objective To observe the improvement effect of Chongcao Baoshen Granules(CCBSG)on renal osteodystrophy(ROD)rats and the influence on Klotho/fibroblast growth factor-23(FGF-23)pathway.Methods A total of 72 rats were randomly separated into control group,ROD group,low dose,medium dose,and high-dose CCBSG groups,and calcitriol group,12 in each group.Except control group,all other groups were used to construct ROD rat models through 5/6 nephrectomy combined with high phosphorus feeding,and after successful modeling,medication was administered once a day for 12 weeks.Automatic biochemical analyzer was applied to detect levels of urea nitrogen(BUN),creatinine(Scr),calcium(Ca2+),and phosphorus(P3+)in serum;ELISA method was applied to detect the level of intact parathyroid hormone(iPTH)in serum;dual energy X-ray was applied to detect bone mineral density of femur;HE staining was used to evaluate the pathological changes of femoral tissue;Western Blot was used to detect Klotho and FGF-23 proteins expression in femoral tissue.Results Compared with control group,the pathological damage of the femur in ROD group was severe,the levels of BUN,Scr,P3+,iPTH in serum and the expression of FGF-23 protein in femoral tissue increased,the level of Ca2+,bone density,and Klotho protein expression in femoral tissue decreased(P<0.05);compared with ROD group,the pathological damage to the femur of rats in low dose,medium dose,and high dose CCBSG groups,and calcitriol group was alleviated,the levels of BUN,Scr,P,iPTH in serum and the expression of FGF-23 protein in femoral tissue decreased,the level of Ca2+,bone density,and the protein expression of Klotho in femoral tissue increased(P<0.05).Conclusion The improvement of rat ROD by CCBSG may be related to the regulation of the Klotho/FGF-23 pathway.
