Correlation analysis between HPLC fingerprint and cell viability of Coptis chinensis
Objective To establish the HPLC fingerprints of Coptis chinensis and study the relationship between its spectrum-effect and cell viability,and clarify the differences in active ingredients and quality of Coptis chinensis from different origins.Methods The fingerprints of Coptis chinensis were established by HPLC method and the similarity was evaluated by combining cluster analysis(CA),principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA)to evaluate the shared peaks.The effect of Coptis chinensis on the cell viability of Hep3B cells was used as the efficacy index,and the spectrum-effect relationship was analyzed by a combination of grey correlation analysis(GRA)and orthogonal partial least squares regression analysis(PLSR).Results HPLC fingerprint profiles of different origins of Coptis chinensis were established,10 common peaks were identified,the similarity of all samples was greater than 0.98,and 8 peaks were attributed by comparison method.The CA results showed that all samples were clustered into 2 categories,which was in general agreement with the PCA results.Combined with orthogonal partial least squares-discriminant analysis(OPLS-DA),it was found that berberine,palmatine and grandisine were the three signature compounds responsible for the differences among the sample groups of Coptis chinensis.The spectroscopic correlations showed that berberine,palmatine,columbamine were the main components that differentiated the inhibition of cell viability in the alcoholic extracts of different origins of Coptis chinensis.Conclusion Different origins of Coptis chinensis are characterized by overall quality consistency and variability of specifiic components,and berberine,palmatine and columbamine are closely related to the inhibition of cell viability,providing a reference basis for the quality evaluation of Coptis chinensis from different origins.
