To study the mechanism of modified Bazheng Powder in the prevention and treatment of calcium oxalate renal calculi in rats based on autophagy regulation
Objective To investigate the mechanism of modified Bazheng Powder in the prevention and treatment of calcium oxalate renal calculi in rats.Methods Twenty-four SPF SD male rats were randomly divided into normal group,model group,modified Bazheng Powder group and potassium sodium hydrogen citrate group,with 6 rats in each group.Calcium oxalate model was established in all the other groups except normal group.At the same time,the general condition and body weight of the rats during drug administration were recorded,and the levels of creatinine(Cr)and urea nitrogen(BUN)in blood,malondialdehyde(MDA)and superoxide dismutase(SOD)in renal tissue of the rats were detected.HE staining and VonKossa staining were used for renal pathological detection,and Western Blot was used to detect the expression levels of autophagy related proteins Beclin1,P62,and LC3 Ⅱ/LC3 Ⅰ in renal tissue.Results Compared with model group,BUN and Cr levels in blood of modified Bazheng Powder and potassium sodium hydrogen citrate groups were significantly decreased(P<0.01),renal tubule dilation was significantly reduced,crystal deposition was significantly reduced,while MDA content in renal tissue was significantly decreased(P<0.01),SOD content increased significantly(P<0.01).Western Blot results showed that,compared with model group,modified Bazheng Powder group could significantly up-regulate Beclinl,LC3 Ⅱ/LC3 Ⅰprotein expression levels(P<0.05).Conclusion Modified Bazheng Powder may reduce oxidative stress and enhance autophagy to induce protective autophagy and clear cell damage in rats with calcium oxalate renal calculi.
