有氧运动联合薰衣草精油对睡眠剥夺大鼠学习记忆及炎症因子水平的影响

目的 观察有氧运动联合薰衣草精油吸嗅干预调节NF-κB信号通路对睡眠剥夺大鼠学习记忆及炎症因子水平的影响.方法 将40只SPF级雄性SD大鼠随机分为5组,分别为空白组、模型组、运动组、精油组、运动联合精油组.运动组行有氧运动,精油组行薰衣草精油吸嗅,运动联合精油组行有氧运动联合薰衣草精油吸嗅,均预干预4周后,模型组、运动组、精油组和运动联合精油组大鼠进行72 h的急性睡眠剥夺.Morris水迷宫检测大鼠的学习记忆能力,HE染色观察海马组织病理形态变化,Western Blot检测大鼠海马中IκBa、AC-NF-κB蛋白水平,实时荧光定量PCR技术和ELISA实验检测大鼠海马和血清中肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)表达水平.结果 与空白组比较,模型组大鼠逃避潜伏期和游泳路程明显延长,初次抵达平台时间明显延长,目标象限停留时间明显缩短(P<0.01);大鼠海马神经元排列混乱,密度减少且数量降低;IKBa蛋白表达显著降低,AC-NF-κB蛋白表达显著增高(P<0.01);海马及血清中炎症因子TNF-α、IL-6、IL-1β表达水平显著上升(P<0.01).与模型组比较,运动组、精油组和运动联合精油组大鼠潜伏期和游泳路程明显缩短,目标象限停留时间延长(P<0.05,P<0.01);大鼠海马神经元排列较为规整,密度增加且数量增高;运动组大鼠AC-NF-κB蛋白表达降低,精油组和运动联合精油组大鼠IκBα蛋白表达显著增高,AC-NF-κB蛋白表达显著降低(P<0.05,P<0.01);海马及血清中炎症因子TNF-α、IL-6、IL-1β的表达量显著降低(P<0.01).结论 有氧运动联合薰衣草精油能够减轻大鼠海马组织及血清中的炎症反应,改善睡眠剥夺引起的大鼠学习记忆能力损伤.
Aerobics exercise combined with lavender essential oil effects the learning memory and inflammtory factor levels of sleep deprived rats
Objective To observe the effects of aerobic exercise combined with lavender essential oil inhalation and sniffing intervention to regulate NF-κB pathways on learning memory and inflammatory factor levels in sleep deprived rats.Methods Forty male Sprague-Dawley rats were divided rapidly into five groups:blank group,model group,exercise group,essential oil group,and exercise combined essential oil group.Exercise group performed aerobic exercise,essential oil group performed lavender essential oil inhalation and sniffing,and exercise combined essential oil group performed aerobic exercise associated with lavender essential oil inhalation and sniffing,all of which were pre-intervened for 4 weeks,and then the rats in model group,exercise group,essential oil group,and exercise combined essential oil group underwent 72 h of acute sleep deprivation.The Morris water maze was developed to test the learning and memory of rats.HE staining was used to monitor pathological morphological changes in hippocampal tissue;Western Blot was performed to detect the expression levels of IκBαand AC-NF-κB proteins in rat hippocampa;real-time quantitative fluorescence PCR and ELISA assay were used to detect the expression amounts of TNF-α,IL-6 and IL-1 β in hippocampa and serum.Results Compared with blank group,model group had considerably longer evasion latency and swimming distance,longer initial arrival time at the platform,and shorter residence time in the target quadrant(P<0.01);rat hippocampal neurons were disorganised,with a reduced density and number;IκBαprotein expression was considerably lower,and AC-NF-κB protein expression was considerably higher(P<0.01);and the expression levels of inflammatory factor TNF-α,IL-6,IL-1 β expression levels increase remarkably(P<0.01).Compared with model group,rats in exercise group,essential oil group and exercise-combined essential oil group had obviously shorter latency and swimming distance,and longer residence time in the target quadrant(P<0.05,P<0.01);hippocampal neurons in the rats were arranged more regularly,with increased density and higher number;AC-NF-κB protein expression in exercise group was reduced,and IκBα protein expression in essential oil group and exercise-combined essential oil group was obviously increased,while AC-NF-κBα protein expression in the hippocampus and serum was obviously increased(P<0.05),while AC-NF-κB protein expression was decreased remarkably(P<0.05,P<0.01);the expression of inflammatory factors TNF-α,IL-6,and IL-iβ in the hippocampus and serum was decreased remarkably(P<0.01).Conclusion Aerobic exercise combined with lavender essential oil can attenuate the inflammatory response in the hippocampus and serum of rats,and ameliorate the learning and memory deficits induced by sleep deprivation.
