Study on the content and quality evaluation of effective ingredients in wine-precessed Scutellaria and its formula granules
Objective To study the content and quality evaluation of effective ingredients in wine-precessed Scutellaria and its formula granules.Methods The content of baicalin,baicalein,and wogonin in 10 batches of wine-precessed Scutellaria samples,as well as the effective components of wine-precessed Scutellaria formula granules were determined by high-performance liquid chromatography.The chromatographic conditions of wine-precessed Scutellaria were:detection wavelength of 218 nm,column temperature of 25℃,flow rate of 1.0 mL/min,and injection volume of 10 μL;the chromatographic conditions of wine-precessed Scutellaria formula granules were:detection wavelength of 280 nm and column temperature of 30 ℃,the flow rate of 0.3mL/min,and the injection volume of 10 μL.Established chromatographic conditions were used for qualitative and quantitative analysis of wine-precessed Scutellaria and its formula granules.Results There was a good linear relationship(r>0.9990)between the three flavonoids in Scutellaria and wine-precessed Scutellaria.The average recovery rates of baicalin,baicalein,and wogonin were 100.15%,101.89%,and 103.18%,respectively(RSD<3.0%).This method has good repeatability,which is easy to operate and stable,and can be used for the content determination of effective ingredients in wine-precessed Scutellaria and its formula granules.The content determination results showed that the content of baicalin in 10 batches of Scutellaria samples was>8.0%and the average content of baicalin in wine-precessed Scutellaria formula granules was 156.66 mg/g,which was within the limited range,calculated as baicalin,ranging from 142 to 265 mg/g.Conclusion The content of effective ingredients in both wine-precessed Scutellaria and its formula granules meets national standards,and the comprehensive quality evaluation is good,providing a basis for the standardization,modernization,and internationalization of Scutellaria medicinal materials in clinical application.
