摘要

肺癌是全球癌症发病率和死亡率的主要原因.顺铂(Cisplatin,CDDP)是临床肺癌治疗方案中普遍使用的药物,但毒副作用较强,且易产生耐药性.环吡酮胺(Ciclopirox olamine,CPX)是一类广谱抗菌药,具有潜在的抗肿瘤效果.文中拟将CDDP与CPX联合研究其抗肿瘤效果.MTT法研究表明,二者联合可显著降低肿瘤细胞的存活率,在 1∶10 的比例下对A549 细胞存在较强的药物协同作用.通过细胞克隆与划痕实验、细胞周期和免疫荧光技术等,证明CDDP联合CPX显著抑制肿瘤细胞的增殖和迁移,使DNA损伤,阻滞细胞周期于G1 期,并通过降低Bcl-2/Bax蛋白比例造成线粒体损伤.结果表明,CDDP联合CPX可协同诱导A549 细胞凋亡,为临床上非小细胞肺癌的联合用药治疗提供新的有效方案.

Abstract

Lung cancer is the primary cause of cancer morbidity and mortality worldwide.Cisplatin(CDDP),a drug commonly used in the clinical treatment of lung cancer,has the disadvantages of strong toxicity and side effects,and is prone to drug resistance.Ciclopirox olamine(CPX),a class of broad-spectrum antibacterial drugs,shows some potential anti-tumor effect.In this study,CDDP and CPX would be combined to study their anti-tumor effect.The MTT assay showed that the combination of CDDP and CPX could significantly reduce the viability of cancer cells,and had a strong synergistic effect on A549 cells at a ratio of 1∶10.The proliferation and migration of cancer cells were significantly inhibited by colony formation assay and wound healing assay;DNA was damaged and the cell cycle was arrested in G1 phase by cell cycle assay;the ratio of Bcl-2/Bax was detected and mitochondria were damaged by western blot analysis.The results show that CDDP combined with CPX can synergistically induce apoptosis of A549 cells,which provides a new and effective solution for the clini-cal combination therapy of non-small cell lung cancer.
