PPM1A抑制胰腺癌细胞PANC-1的迁移侵袭和EMT进程

为研究镁依赖性蛋白磷酸酶 1A(protein phosphatase magnesium-dependent 1A,PPM1 A)对胰腺癌细胞迁移、侵袭及上皮-间充质转化(epithelial-mesenchymal transformation,EMT)的影响,采用生物信息学技术以及细胞免疫荧光实验分析PPM1A在胰腺癌中的表达情况,划痕、Transwell、qRT-PCR、Western blot、细胞免疫荧光实验检测PPM1A对胰腺癌细胞PANC-1迁移、侵袭及EMT标志物表达水平的影响,生物信息学网站预测PPM1A的上游miRNA.结果表明,PPM1A在胰腺癌中低表达,抑制PANC-1细胞的迁移、侵袭和EMT进程,miR-105-5p为PPM1A的潜在上游miRNA.说明PPM1A和miR-105-5p可能是治疗胰腺癌新的靶点.
PPM1A inhibits the migration,invasion and EMT process of PANC-1 pancreatic cancer cells
To investigate the effects of protein phosphatase magnesium-dependent 1A(PPM1A)on the migration,invasion and epithelial-mesenchymal transformation(EMT)in pancreatic cancer(PC),bioinformatics analysis and cellular immunofluorescence methods were used to study the expression of PPM1A.Wound healing,Transwell,qRT-PCR,Western blot and immunofluorescence tests were used to detect the effects of PPM1A on the migration and invasion of PANC-1 cells and the expression level of EMT markers.The bioinformatics website predicts upstream miRNAs of PPM1A.The results showed that PPM1A was underexpressed in PC and inhibited the migration,invasion and EMT process of PANC-1 cells,miR-105-5p was a potential upstream miRNA of PPM1A,and PPM1A and miR-105-5p may be new targets for the treatment of pancreatic cancer.
