Effects of ODC1 downregulation on the proliferation,apoptosis,and sensitivity to platinum drugs of human SCLC cells H82 and H69
To explore the effect of the downregulation of ornithine decarboxylase1(ODC1)on the proliferation,apoptosis and platinum drugs sensitivity of human small cell lung cancer(SCLC)cells H82 and H69,the expression levels of ODC1 of the human SCLC cell lines were examined.H82 and H69 cells were infected with the LV-NC/ODC1-RNAi lentivirus,then the expression of ODC1 at the mRNA and protein levels was determined by real-time fluorescent quantitative PCR(q-PCR)and immunoblot assay(Western blot).CCK-8 assay,colony formation assay,flow cytometry and Western blot were used to detect the changes of cell proliferation,apoptosis and sensitivity to platinum drugs after downregulation of ODC1.As a result,ODC1 was mostly highly expressed in human SCLC cell lines;the proliferation of human SCLC cells H82 and H69 slowed down;their apoptosis significantly increased and drugs sensitivity was enhanced.In summary,downregulation of ODC1 can inhibits the proliferation of human SCLC cells H82 and H69,promotes cell apoptosis,and enhances cell sensitivity to platinum drugs.
