摘要

为明确饲料中添加雨生红球藻(Haematococcus pluvialis)对克氏原螯虾(Procambarus clarkii)肝胰腺基因表达的影响,以克氏原螯虾商业饲料作为基础饲料添加雨生红球藻,配置含有3‰雨生红球藻的试验饲料对克氏原螯虾进行为期45 d的喂养.基于RNA-seq技术对其肝胰腺的转录组数据进行分析,测序共获得44.61 Gb干净数据,Q30碱基百分比在92.46%以上.在FDR≤0.05且|log2FC |≥3的筛选条件下转录组测序分析共鉴定出895个差异表达基因.GO功能富集分析结果显示,差异表达基因主要在能量代谢过程上显著富集.KEGG通路富集分析结果显示,差异基因主要在能量代谢、免疫和神经传递相关通路中被显著富集.综上所述,饲料中添加3‰的雨生红球藻可能有助于克氏原螯虾提高抵抗病原体和外界环境变化的能力.

Abstract

To investigate the impact of Haematococcus pluvialis supplementation on hepatopancreas gene expression in Procambarus clarkii,commercial feed for Procambarus clarkii was used as the base diet and supplemented with Haematococcus pluvialis at a concentration of 3‰.The experimental group was fed with modified diet for 45 days,and RNA-seq technology was employed to analyze the transcriptome data from the hepatopancreas.A total of 44.61 Gb clean sequencing data were ob-tained,with over 92.46% Q30 bases.Through transcriptome sequencing analysis using screening cri-teria(FDR≤0.05 and |log2FC |≥3),we identified 895 differentially expressed genes.GO functional enrichment analysis revealed that these differentially expressed genes were primarily associated with energy metabolism pathways.KEGG pathway enrichment analysis demonstrated significant enrich-ment of differential genes in energy metabolism,immunity,and neurotransmission-related path-ways.In conclusion,supplementing Procambarus clarkiis diet with 3%o Haematococcus pluvialis may enhance its resistance against pathogens and environmental changes externally imposed upon it.
