一株饲料鳜源魏斯氏菌的分离鉴定及其抑菌特性

为筛选具有广谱抑菌性的益生菌用于饲料鳜鱼养殖,分离定植于摄食人工配合饲料的鳜(Siniperca chuatsi)肠道优势菌并进行体外抑菌特性研究.从饲料鳜肠道中分离、培养出1株优势益生菌(命名:SJ548),对该菌进行形态学、生理生化以及16S rDNA分子鉴定,使用牛津杯法测定该优势菌对8种指示菌的抑菌活性.菌株SJ548在MRS固体培养基上呈现白色、圆形、边缘圆润的菌落,染色镜检为杆状或双杆状的革兰阳性菌;通过生理生化测试及16S rDNA基因序列分析,将菌株SJ548鉴定为食窦魏斯氏菌(Weissella cibaria).研究发现:菌株SJ548可以抑制大肠杆菌(Escherichia coli)、嗜水气单胞菌(Aeromonas hydrophila)、金黄色葡萄球菌(Staphylococcus aureus)、沙门氏菌(Salmonella enterica)、鳗弧菌(Vibrio anguillarum)、哈维氏菌(V.harveyi)、副溶血弧菌(V.parahaemolyticus)、溶藻弧菌(V.alginolyticus)的生长,抑菌圈的直径为13~18 mm.本试验从饲料鳜肠道中分离获得的SJ548作为益生菌,可作为候选菌株开发为益生菌添加剂.
Isolation,identification and antibacterial characteristics of a Weissella strain from feed mandarin fish(Siniperca chuatsi)
In order to select probiotics with broad-spectrum bacteriostatic properties for use in the cul-ture of mandarin fish(Siniperca chuatsi),the dominant bacteria was isolated from the intestine of mandarin fish,which ingests artificial diets,and their bacteriostatic properties were investigated in vitro.A dominant probiotic strain(SJ548)was isolated and cultured from the intestine of feed manda-rin fish.It was identified morphologically,physiologically,biochemically,and molecularly by 16S rD-NA,and the inhibitory activities of SJ548 were determined by an Oxford Cup assay on 8 indicator or-ganisms.SJ548 showed white,rounded colonies with rounded edges on MRS solid medium,and stai-ning microscopy showed that it was a Gram-positive bacterium with rod-shaped or double rods.Through physiological and biochemical tests and 16S rDNA gene sequence analysis,SJ548 was identified as a Weissella cibaria.It was found that strain SJ548 could inhibit Escherichia coli,Aeromonas hydrophila,Staphylococcus aureus,Salmonella enterica,Vibrio anguillarum,V.harveyi,V.parahaemolyticus,and V.alginolyticus,and the diameter of the circle of inhibition was 13~18 mm.SJ548 isolated from the intestinal tract of feed mandarin fish in the present experiment can be used as a candidate strain for development as a probiotic,which can provide a scientific basis for the research and development of artificial feed additives for mandarin fish and the ecological culture of mandarin fish.
