pH、葡萄糖和几种阳离子对本溪细鳞鱼精子活力的影响

为给本溪细鳞鱼(Brachymystax lenok Pallas)人工繁殖中最佳受精激活介质的选择提供理论依据,也为细鳞鱼的精子稀释液和保存液的配制提供科学依据,采用平板载片法,研究了 pH、葡萄糖和Na+、K+、Ca2+、Mg2+等金属离子以及冷藏时间对本溪细鳞鱼精子活力的影响.结果显示,精子应在超低温条件下进行保存.研究表明,应采用自来水配置,pH为8,且应含有0.5 g/L Na+、0.3 g/L Mg2+、0.3 g/L Ca2+和50 mmol/L葡萄糖,从而达到本溪细鳞鱼人工授精的最佳条件.研究得出,适宜浓度的pH值和葡萄糖、Na+、Mg2+、Ca2+对本溪细鳞鱼精子活力有增强作用.K+会抑制本溪细鳞鱼的精子活力,而Na+是促进本溪细鳞鱼精子活力的主要因素.
Effects of pH,glucose and several cations on sperm motility of Benxi Brachymystax lenok Pallas
To provide a theoretical basis for the selection of the best fertilization activation medium in the artificial breeding of Benxi Manchurian trout(Brachymystax lenok Pallas),and also to provide a scientific basis for the preparation of sperm dilution and preservation solution of the fish,the effects of pH,glucose,metal ions such as Na+,K+,Ca2+,Mg2+and cold storage time on sperm motility of Benxi Manchurian trout were studied by plate slide method.The results showed that sperm should be stored at ultra-low temperatures,the activation solution of artificial insemination of Benxi Manchuri-an trout should be prepared with tap water,pH 8,and contain 0.5 g/L Na+,0.3 g/L Mg2+,0.3 g/L Ca21and 50 mmol/L glucose.It was concluded that the appropriate pH value and glucose,Na+,Mg2+,Ca2+can enhance sperm motility of Benxi Manchurian trout.K+can inhibit the sperm motility of Benxi Manchurian trout.Na+is the main factor promoting sperm motility of Benxi Manchurian trout.
