Methods of improving inoculum quality of vitamin C two-step fermentation
The effects of L-cysteine or(and)calcium chloride on the growth and production of 2-keto-L-gulonic acid(2-KLG)in two-step culture medium of vitamin C were studied.The results showed that adding appropriate amount of L-cysteine and(or)calcium chloride to the two-step culture medium could promote the growth of Ketogulonigenium vulgare,increase the cell density,coordinate the mixed bacteria proportion and improve the quality of the mixed strains,the production rate of 2-keto-L-gulonic acid was increased and the fermentation period was shortened.The effect of adding 0.3 g/L cysteine and 0.6 g/L calcium chloride in the seed liquor culture medium was the most remarkable.The concentration of Ketogulonigenium vulgare in the seed liquor increased by 30.77%,and the fermentation period shortened by 8.33%,the formation rate of 2-keto-L-gulonic acid increased by 9.31%.
