Effects of hydroxyapatite coating on osseointegration of polycaprolactone screws prepared by polydopamine biomimetic method after anterior cruci-ate ligament reconstruction
Aim:To prepare hydroxyapatite(HA)coating on polycaprolactone(PCL)screws by polydopamine biomim-etic method,and evaluate the effects on osseointegration in anterior cruciate ligament reconstruction.Methods:The porous PCL screw was designed and printed by 3D printer.HA coating was prepared by polydopamine biomimetic method.Twenty adult male New Zealand white rabbits were selected to establish the model of anterior cruciate ligament reconstruction in the bilateral hindlimbs and randomly allocated into 2 groups,and PCL screws with HA coating(experimental group)and without HA coating(control group)were implanted.Ten rabbits were killed 6 and 12 weeks after operation,and the specimens of fe-mur-anterior cruciate ligament-tibia complex were prepared.The osseointegration of screw bone interface was evaluated by Masson staining,Micro-CT and biomechanical test.Results:Masson staining showed that at 6 and 12 weeks after operation,more new bone trabeculae formed around the material and grew into the pores of the material,but the amount and depth of new bone trabeculae in the experimental group were better than those in control group.Micro-CT showed that the new bone in the material interface of the 2 groups gradually increased at 6 and 12 weeks after operation,and the new bone mass a-round the experimental group was more than that of control group.Compared with 6 weeks after operation,the bone volume fraction,trabecular number and bone mineral density increased,while the trabecular separation rate decreased in both groups at 12 weeks after operation,and the above indexes in the experimental group were better than those in control group at 6 and 12 weeks after operation(P<0.05).At6 and12 weeks after operation,the peak tension of the experimental group was higher than that of control group[(39.98±5.32)N vs(27.46±6.95)N,(62.64±8.76)N vs(50.15±6.48)N](P<0.05).Conclusion:The HA coating of PCL screws prepared by polydopamine biomimetic method can promote os-seointegration after anterior cruciate ligament reconstruction and improve the biomechanical stability of interface screws.
